THIADIAZOLE DERIVATIVES: A COMPREHENSIVE SURVEY OF SYNTHESIS APPROACHES

Thiadiazoles are an important class of heterocyclic compounds that have attracted significant attention due to their wide spectrum of biological and pharmacological activities, including antimicrobial, anti-inflammatory, anticancer, and antitubercular properties. Their unique heteroatom-rich structure confers high reactivity, making them versatile scaffolds in medicinal and synthetic chemistry. This review provides a comprehensive overview of the synthetic strategies employed for the preparation of thiadiazole derivatives, with emphasis on conventional chemical routes, cyclization methods, and modern approaches such as green chemistry and catalytic techniques. The advantages and limitations of different methods are highlighted, alongside their relevance to pharmaceutical applications. The study also discusses recent advances in functionalization strategies aimed at enhancing bioactivity and selectivity. Overall, this survey emphasizes the importance of thiadiazole synthesis in advancing drug discovery and material sciences
